Overview of Centrosomal Genes

Centrosome is the main microtubule organizing center (MTOC) in most animal cells, which is of great importance for fundamental cellular functions, such as the formation of bipolar spindles, cell division during mitosis, organizing microtubules and transporting mitochondria to their functional destinations. Genes that encoding proteins forming centrosome structure or localized in pericentromeric material (PCM) are called centrosomal genes, which play very important role in many biological processes. For example, overexpression of Aurora B results in multinucleate cells, and the overexpression of Plk4 causes centrosome amplification, which leads to tumorigenesis. Ploidy alteration is one of the most common mechanisms of centrosome anomalies in cancer, and multiple stimuli induce polyploidy and centrosome aberrations, including the inactivation of BRCA1, BRCA2, SKP2, and GADD45 genes. Therefore, the proteins encoded by the centrosomal genes are of great value as potential therapeutic targets of cancers.

Applications of Centrosomal Gene Editing

Centrosomal gene editing animal models are valuable for functional studies of centrosomal genes, as well as therapeutic development of centrosome-related diseases, especially cancers. 3% of all human protein-coding genes encode proteins localized to the centrosome or the centriolar satellites. And Gene Ontology (GO)-based analysis of genes encoding proteins that localize to centrosomes or centriolar satellites shows the enrichment of terms are related to organization of the microtubule cytoskeleton, ciliogenesis, cell cycle progression, and organization of organelles.
