Actin is the most abundant protein in most eukaryotic cells. The ability of actin to shift between monomeric (G-actin) and filamentous (F-actin) states under the control of nucleotide hydrolysis, ions and a large number of actin-binding proteins gives actin a key role in many fields.
Vertebrates express three major actin isoforms, including three α-isoforms in skeletal, cardiac, and smooth muscle and β and γ isoforms expressed in non-muscle and muscle cells.
α-Actin | α-actin consists of 377 amino acids, has a molecular weight of about 42 kDa, and is widely distributed in almost all muscle-type cells. This protein is mainly used to detect skeletal muscle, smooth muscle, vascular smooth muscle, cardiac muscle and myogenic tumors. |
β-Actin | β-actin is composed of 375 amino acids and has a molecular weight of approximately 42 kDa. Antibodies to β-actin are mainly used to label smooth muscle and tumors of its origin. β-actin protein is usually expressed at unchanged levels in cells and is therefore widely used as a reference for consistent loading of samples at Western, and is also often used for immunostaining to observe the microfilament structure of cells. |
γ-Actin | There is a link between γ-actin and microtubules. Inhibition of γ-actin expression leads to impaired cell migration, motility and polarity. |
