Lysine-based VLPs Modifications
The classical approach to form cVLPs by chemical conjugation is based on lysine modification-using a cross-linking agent that reacts with the nucleophilic amino acid side chain on the VLPs. Because lysine has a primary amine, it is easily modified. When the pH is between 7.2 and 9, the lysine residues exposed on the surface form amide bonds through the ester reaction of n-hydroxysuccinimide (NHS) (NHS can be released as part of the reaction).

Application of Lysine-based Modifications of VLPs
This lysine-based chemical conjugation modification has been used to embed transferrin in VLPs, allowing VLPs to cross the blood-brain barrier and opening up a new therapeutic avenue for the treatment of neurological disorders. In addition, peptides, proteins and fluorescent probes can also be embedded in VLPs via the NHS reaction, resulting in cVLPs.
