Overview of Centrosome Localization

Centrosome is the major microtubule organizing centers (MTOCs) in most cells. Microtubules (MTs) are important cytoskeletal structures, essential for cell division, intracellular transport, motion of cilia and flagella, as well as cell migration and establishment of polarity. During the mitosis, centrosome duplicate once at the beginning of G1 phase and each of them is localized in each pole of the spindle for maintaining the cell bipolarity. By these activities, centrosome is involved regulating polarized cell behaviors, such as cell migration or neurite outgrowth, as well as mitotic spindle orientation.

Localization of Centrosomes in Cell Migration

In slow-moving cell, the centrosome is usually located in front of the nucleus (relative to the direction of cell migration) and MTs radiate primarily towards the leading edge. In fast-moving cell, centrosome and MTs localize behind the nucleus during directional migration.

Methods for Centrosome Localization

Live-cell imaging is an important analytical tool in biological research, which provides the subcellular events and biological processes. With the variety of fluorescent proteins and multicolored probes that have been developed, it is now possible to label virtually any molecule. Centrosome is localized by placing genetically encoded fluorescent protein (FP) tags at the N- or C-termini of centrosome proteins, such as tubulin, PCM proteins or CEP proteins.
