Overview of Centrosome Isolation

Centrosome is a cytoplasmic organelle plays important role in many biological processes, such as mitosis, assembling spindle and cell division, as well as cell migration, invasion and polarity. Centrosome is consisted of a pair of cylindrical structures known as centrioles, decorated at their distal ends by appendage structures, and surrounded by a proteinaceous matrix known as the pericentriolar material (PCM). Due to the high-association between centrosome abnormality and malignant tumor, it's valuable to study the spatial and temporal dynamics of centrosome composition and its variation between different cells and tissue.

Glycerol Cushion Sedimentation

Glycerol Cushion Sedimentation

Animal tissues, culture animal cells, or plant tissue are harvested. After preparation of a cytoplasmic extract, Taxol was added to induce microtubule polymerization. Microtubules and centrosomal mRNAs were purified by sedimentation through a glycerol cushion.

Sucrose Density Gradient Centrifugation

Sucrose Density Gradient Centrifugation

To isolate pure centrosome from cultured cells or animal tissues, sucrose gradient centrifugation will be conducted. A low ionic strength buffer can help centrosome detach from nuclei and intermediate filaments. After aggregation and centrifugation, chromatin can be removed, and centrosomes are sedimented onto a sucrose cushion and further enriched by sucrose gradient centrifugation.

Affinity Capture

This method involves a single-step affinity purification of centrosomes by selecting proteins known to localize to distinct sites of the centrosome. And they are immunoprecipitated from induced cells, and reproducibly determined their interactome with mass spectrometry.
