PPI Studies of Centrosomal Proteins

As the major microtubule-organizing center (MTOC) of cell, centrosome functions in various of cellular processes including cell migration, neuronal path finding, axon selection, immune cell function, cell polarity and mitosis. The centrosome consists of a pair of centrioles and the pericentriolar material (PMC). Centrioles are composed of 9 triplet microtubules (MTs) and many associated proteins arranged as a barrel.

Many PPIs of centrosomal proteins have been identified, which facilitated the understanding of centrosomal biology processes. For example, the interactions between Sas6, STIL/Ana2/Sas5, Cep135/Bld10 and CPAP/Sas4 are identified as one of the earliest events in the construction of centrioles and the 9-fold symmetry of MTs. However, there are still a lot of challenges in understanding the PPIs of centrosomal proteins, such as its micro-scale size, then complex composition of proteins and the unstable state of most centrosomal proteins. Therefore, there is a great need for technologies that would uncover the potential PPIs of centrosomal proteins in a quick and high-throughput manner.
