Introduction into Centrosomal mRNA Imaging

Centrosome is the major microtubule organizing center (MTOC) in proliferating animal cells, which is composed with one pair of cylindrically shaped centrioles surrounded by pericentriolar materials (PCMs) including proteins and mRNAs. Centrosome abnormalities cause mis-segregation of chromosome during mitosis, which triggered malignancy. Centrosome amplification (CA) is a hallmark of cancers. Mechanistically, CA compromises fidelity of mitosis and leads to chromosome instability (CIN), which is the basis of tumor initiation and progression.

Due to the development of microscopy and image analysis technologies, detection and quantification CA are easier than ever. CA is caused by several events, and the accumulation of PCMs is included. Therefore, to investigate how centrosomal mRNAs are spatially distributed with respect to spindle microtubules in vivo during cell mitosis and the enrichment on microtubules in different stages even cell types, CD BioSciences provides mRNA imaging services.

FISH (fluorescence in situ hybridization) is a new in situ hybridization method formed by directly using fluorophore labeling or indirect biotin labeling instead of isotopic labeling, which enables relative qualitative and localization analysis of target nucleic acid sequences. FISH has been widely used in various fields of biomedical science because of its safety, rapid, sensitivity, long probe retention time and simultaneous display of multiple colors.
